Spouses looking for silver in Mexico find a 300 year old severed hand driven by a demon.

A British woman visits her husband at a Mexican mine that he is attempting to reopen and discovers that the workers refuse to enter the mine because of an ancient curse. The couple enters the mine to prove there is no danger and inadvertently releases a demon that possess people's left hands and forces them to behave in a suitably diabolical manner. The only way for a possessed person to free themselves from this torment is to cut off his/her left hand, after which it scurries away in search of its next victim.

In an underground chamber, hooded devil worshipers slice the left hand off a young woman, which crawls on its own until a cultist stabs it and places it in a silver case.

Three hundred years later.

Jennifer Baines (Samantha Eggar) arrives in Guanajuato, Mexico, where her husband Mark Baines (Roy Cameron Jensen) is reopening a mine. Inside, Jennifer accidentally causes a small rock slide which reveals a mummified corpse with no left hand, and the workers fear the mine is cursed by the "devil's hand." Hoping to inspire the workers to return to work, Jennifer and Mark go back inside. Upon reaching the lowest level of the mine, they discover the deserted chamber and Mark finds the silver hand-shaped case. He shows it to the workers, announcing the devil's curse is broken, but the workers retreat in fear.

That night, after Jennifer goes to bed, an intoxicated Mark opens the case and empties the ashy contents onto the table. As he staggers to bed, the ashes re-assemble into a hand, crawl into the bedroom and up Jennifer's leg. She awakens screaming and Mark fights with the hand until it disappears into his left hand, and he runs out of the room.

The next morning, Jennifer searches for Mark and discovers that he has ordered the workers back into the mine, rigging the site with explosives. Despite her protests, Mark blows up the mine and drives away. Later, Jennifer sees a newspaper headline announcing Mark is a success at winning a lot of money, gambling at the Sands hotel in Las Vegas, Nevada.

At the hotel, a gambler orders his girl friend, Angela, to flirt with Mark. As Jennifer arrives at the front desk, the gambler and Angela abduct Mark and take him to a remote shack. They demand that Mark reveal his gambling secrets, threatening to cut off his hands. Mark's left hand, however, assumes a mind of its own and kills them. Mark tries to cut off his hand, but it douses him with gasoline and sets him on fire.

Later, at the police station, Jennifer learns that three corpses were found at the shack and she is certain the burned man is Mark. The police, however, had assumed the person was the owner of a neighboring shack, and a relative identified the charred corpse before shipping it to Los Angeles, California, for burial.

At night, Jennifer visits the California church where the corpse was sent and questions Father Cunningham (Stuart Whitman), who presided over the burial. He leads her to the grave, but before they arrive, the body exhumes itself and hides. Father Cunningham thinks the grave was vandalized and does not believe Jennifer's contention that the body dug its way out. He calls Sgt. Leo Matson (Lew Saunders) to investigate and while they wait, Jennifer shows him the silver hand-case, but the priest doubts her tale of the devil's crawling hand.

Meanwhile, Matson checks the grave as the corpse sneaks to the police car and slams the door on its left hand, severing it. When Matson returns to the vehicle, he finds the collapsed corpse and is attacked by the detached hand. Flashes of lightning bring Jennifer and Cunningham outside as Matson speeds away, leaving behind the corpse with no left hand.

The next day, at a boxing gym, Cunningham questions Matson who claims he was called to another crime scene. Mattson offers to box with the priest and attacks his opponent with his left hand until Cunningham's crucifix slips outside his shirt, and Matson runs away.

Back at the church, Cunningham claims Matson is not supernaturally possessed, but Jennifer disagrees and leaves the address of her hotel, announcing she is staying in Los Angeles until the hand is destroyed. As she places her purse in the rental car, Jennifer is confronted by Matson who arrests her for car theft. Instead of bringing her to the police station, he drives her to a medical office, where he handcuffs her to a chair and orders Dr. Julian Rivkin and Nurse Morgan to cut off his left hand, threatening them with death. Revealing that he has been possessed by the "devil's hand," Matson explains the hand is a gift for Jennifer; she set the hand free at the mine, so now it belongs to her. After Dr. Rivkin removes the hand, it reaches for a gun and shoots Nurse Morgan, then flies onto Matson and crushes his face. As the doctor tries to save Matson, he, too, becomes possessed by the hand. Jennifer tries to escape but the doctor sedates her with a syringe full of medication and carries her back into the exam room.

Meanwhile, Father Cunningham notices her car outside the church and finds the silver case inside. When the priest and Patrolman Yates also see Matson's patrol car at the medical office, Cunningham rushes inside as the doctor prepares to cut off his hand as a gift for Jennifer. Frightened away, Dr. Rivkin drives off while Cunningham frees Jennifer. They chase the doctor in Patrolman Yates's car until Rivkin jumps from his vehicle onto the side of a train. As the doctor falls off the train, his left arm lands on the tracks; the train runs over the arm and the severed hand escapes.

Later, Father Cunningham delivers Jennifer to the hotel and offers to help her destroy the hand. As he drives away, he comes upon a car accident and is asked to perform the last rites. When Cunningham notices the victim's left hand missing, he rushes back to the hotel and arrives as Jennifer pulls back the bed covers to find the hand waiting to attack. They escape in Cunningham's car but, as they speed away, the hand jumps from the hotel's second floor onto the vehicle.

At the church, Cunningham attempts to call the police but the phones are dead. The power suddenly goes out and the doors lock on their own. As Cunningham gets candles, the hand attacks him. An iron gate blocks Jennifer's path, so she reaches her left hand through the bars and asks the devil's hand to take her, not Cunningham. The hand moves toward her but Cunningham grabs it and the devil's hand possesses him. A chase ensues until Cunningham finds Jennifer hiding, with a knife, on the opposite side of an iron doorway. When the priest shoves his left hand through the grate toward her, Jennifer offers him the knife, then kisses his hand. As she pulls back, Cunningham stabs the knife through his hand, trapping it in the grating. Jennifer hands him a nearby blow torch and he burns the hand while quoting the scripture: "If your hand offends you, cut it off and cast it aside." The next day, Jennifer and Cunningham scatter the hand's ashes into the ocean and the priest claims the dust will rest with no hope of resurrection.

Later, at Jennifer's home, a deliveryman arrives with a package for her. After the deliveryman leaves, she notices puddles of water and a piece of seaweed on the floor. She cautiously unwraps the box and finds another piece of seaweed with a black candle inside the box. Water drips from a faucet as the devil hand climbs out of the sink and attacks her. The black candle lights itself as the hand throws Jennifer into a glass coffee table, killing her.
